Glutathione is the most abundant intracellular antioxidant in mammalian cells, present at millimolar concentrations. Unlike dietary antioxidants, it is endogenously synthesized and can regenerate other antioxidants (vitamins C and E) after they become oxidized. It also directly neutralizes hydrogen peroxide and lipid peroxides via glutathione peroxidase, and is required by glutathione S-transferases for detoxification of xenobiotics and reactive metabolites.
In melanocyte cell culture models, glutathione inhibits tyrosinase — the rate-limiting enzyme in melanin synthesis — and shifts the balance from dark eumelanin toward lighter phaeomelanin production. This mechanism has been studied in clinical trials investigating hyperpigmentation and photoprotection, making it a frequently used compound in dermatological and cosmeceutical research protocols.
